Description
Chicken embryo extract is prepared from 9-12 day old eggs collected from registered flocks and found to be free from clinical signs of notifiable diseases.
It is processed as a 50% extract in phosphate-buffered saline that is supplemented with 100IU/mL Penicillin, 100 ug/mL Streptomycin and 2.5ug/mL Amphotericin. The liquid extract is sterile filtered to 0.2 µm and lyophilized in 10mL aliquots. The sterility of this lyophilized powder cannot be guaranteed and should be sterile filtered once more after reconstitution.
Lyophilized for longest shelf life.
Preparation and Use
Reconstitute lyophilized product with 10 mL of sterile tissue culture grade water. Agitate to mix. If precipitates appear, continue to agitate under cool conditions for several hours/overnight or sonicate briefly using a sonicating water bath. Then, centrifuge at 3,000 x g for 10 minutes and remove pellet. After reconstitution, lyophilized product must be filtered through a 0.2 µm filter to maintain sterility. Once reconstituted, store at 4°C for immediate use, or aliquot into working volumes and store at -20 to -10°C for long term storage and minimize freeze/thaw cycles. Protein precipitate may be observed on thawing. To remove this, one can centrifuge at 3,000 x g for 10 minutes. Typically used at a concentration of 0.1% to 2% in cell culture.
